Figure 1. The affinity of MonoRab? Rabbit Anti-Humanized VHH Antibody, mAb with different humanized VHHs was tested using ELISA. The coating antigens in separate ELISA assays were Humanized VHHs at 1 μg/mL concentration. The primary antibody was MonoRab? Rabbit Anti-Humanized VHH Antibody, mAb (GenScript, A02165) used at dilutions starting from 1 μg/mL concentration. The secondary antibody was Mouse Anti-Rabbit IgG Fc Antibody (14H9H10)[HRP], mAb (GenScript,A01856), at 1:20,000 concentration.

A camelid single-domain antibody (sdAb, called Nanobody by Ablynx) is a
peptide chain about 110 amino acids long comprising one variable domain of a heavy-chain antibody (VHH fragment). Like a
whole antibody, a single-domain antibody is able to bind selectively to a
specific antigen. Such antibodies allow a broad range of applications in
biotechnical as well as therapeutic fields due to their small size, simple
production, and high affinity. However, their sequences may increase the risk
of immunogenicity and cause anti-drug antibody (ADA) development in humans, and
thus, sdAbs are routinely humanized during development. |
